I am trying to make a higher voltage USB digital potentiometer using the Pololu USB-to-serial adapter and a AD5290 (see illustration below) and was wondering if someone could help me with the wiring. Thanks in advance for any help.

I am not familiar with the AD5290, but from its product page on the Analog Devices site, it looks like it only accepts 3-wire serial or SPI signals. Our Pololu USB-to-Serial Adapter only outputs asynchronous TTL serial signals (UART), so I don’t think you would be able to communicate directly between it and the AD5290.
